This project tests a tool developed within previous Escher research – the Multiple Parameter Risk Response Outcome – in wider populations and for more medicines. Currently, medicines are developed and registered on the basis two things: medicine efficacy, which is based on a single on-target marker and medicine safety, which is based on different biomarkers. Escher research shows that a medicine’s efficacy could also be measured using multiple off-target biomarkers.

In these other Escher studies, a new efficacy algorithm has been developed and validated: the Multiple Parameter Risk Response Outcome or PRO-tool. This tool combines changes in on- and off-target risk markers to predict medicine efficacy. However, previous studies only assessed whether the PRO-tool is applicable to certain kinds of populations and whether the PRO-tool can be used with certain medicines. This study assesses whether the PRO-tool can also be used in populations other than those that have previously been tested; and whether it is effective in predicting medicine efficacy for multiple medicines. The results of this analysis have substantiated the Multiple PRO Tool for predicting the efficacy of an antihypertensive therapy (ACE-inhibitor – fosinopril) and a cholesterol lowering therapy (statin – pravastatin) in a general population cohort. As such, they provide important input for the Escher PREscore Project.
